Glycine is a common exception because it has no corresponding chiral center. D/L naming also cannot simply be equated with writing the entire peptide backwards.

Replacing the D/L configuration at just one position of a peptide and changing the entire peptide into its complete mirror form are also not exactly the same situation analytically.

Why doesn't a mass match answer all the questions?

Changing only the D/L configuration of a certain amino acid can leave the composition and theoretical mass unchanged. Therefore, a matching intact molecular mass cannot by itself prove that the configuration at the required position is correct.

Whether ordinary non-chiral separation can distinguish them depends on the specific molecule and method. Fully mirror-image materials and diastereomeric materials formed by changing some positions should also be discussed separately; one cannot say that all chromatography or all mass spectrometry can always distinguish them or can never distinguish them.

How can additional evidence be sought?

Clearly raise the configuration question with the testing party, then discuss suitable methods and reference standards. This may involve chiral chromatography, chiral amino acid analysis after appropriate treatment, or other methods validated for the specific material.

Some studies use specific tandem mass spectrometry conditions to distinguish diastereomeric peptides containing D-amino acids. This shows that a specific method can provide information, not that routine LC-MS mass results have already completed all chiral confirmation.

The treatment process itself must also be included in the check

If the method requires hydrolysis or other treatment, it is also necessary to assess whether the treatment introduces configurational changes. The final method must be able to answer the question about the original material, not merely measure some property of the treated product.

When ordering, write the D positions, terminal modifications, and full sequence clearly. Labeling only "D peptide" can easily omit which positions need to be D and whether reverse order is also required.

How should an accurate testing request be written?

You can write: We wish to confirm the D/L configuration at specific positions in the given sequence; please state the analytical evidence that can be provided, the methods used, reference standards, and method limitations. This is closer to the identity that actually needs to be confirmed than "a correct molecular weight is enough."

Three questions, three types of evidence needs

Whether the mass matches the expected compositionIs intact mass alone enough: Intact mass can provide information, but it needs to be combined with other identity records

Whether the amino acid sequence is correctIs intact mass alone enough: Further sequence evidence is usually needed

Whether the specified position is DIs intact mass alone enough: Applicable evidence addressing the configuration question is needed

Teaching summary.
