A UV detector then measures how much light they absorb at a selected wavelength. That wavelength is part of the method, not a decorative number on the report. Changing it can change which components are easy to see.

What do 214 nm and 280 nm mean?

The numbers are light wavelengths, measured in nanometres. Thermo Fisher’s peptide-analysis discussion describes detection around 214 nm using peptide UV absorbance. It is one common approach, not a universal requirement for every peptide or every impurity. [1]

Thymosin alpha-1 shows why the choice matters

In its application note on synthetic thymosin alpha-1, Thermo Fisher used UV detection at 214 nm and explained that this peptide has no aromatic amino acids and no UV absorption at 280 nm in that discussion. The example shows why a familiar protein-detection wavelength may be unsuitable for a particular peptide. It is that provider’s method example, not an NHD batch result. [2]

A percentage still depends on what was measured

For a hypothetical calculation, a main-peak area of 980 units out of 1,000 included area units gives 98%. That is an area ratio at the chosen detector settings. It cannot describe a component that the method does not detect or separate. A different response from an impurity can also affect how its area relates to its mass.

Compare methods before comparing headline numbers

Two reports labeled “HPLC purity” may use different wavelengths, columns, gradients or integration rules. A difference in percentage is therefore a reason to inspect the methods, not immediate proof that one supplier has better material. Ask which peaks were included and whether relevant impurities can be resolved.

Ask for the method details with the product

For a thymosin alpha-1 enquiry, specify the molecular form and quantity, then ask which purity method and supporting chromatogram could be provided for the proposed material. Check the NHD product family and HPLC guide together. The product page alone does not confirm a particular batch has been tested.

Useful links

Thymosin alpha-1 product family

HPLC purity guide

Ask about product and chromatogram records

For laboratory research, development and manufacturing only.
