A study on SS-31 offers another way of thinking: when the amount stays the same, function may still change.

TFP deficiency can be understood as a problem in the fat utilization step

Mitochondria must process long-chain fats step by step before they can be used for energy. Mitochondrial trifunctional protein, or TFP for short, takes part in this process. Genetic defects may make the related steps run poorly.

The study used beta-TFP-deficient mice and patient-derived fibroblasts to observe whether SS-31, also called elamipretide, changes mitochondrial function. It did not directly conduct an efficacy trial in patients.

Where is cardiolipin?

Cardiolipin is a lipid in the inner mitochondrial membrane and is related to membrane structure and the working environment of energy-related proteins. It is not a fat exclusive to the heart; the word heart in its name can easily cause misunderstanding.[1]

SS-31 can interact with cardiolipin. The study therefore looked at both energy utilization performance and the total amount and composition of cardiolipin, to see whether functional changes were accompanied by changes in the amount of material.

What combination of results appeared this time?

After treatment, the deficient mice showed improved exercise endurance, and some liver mitochondrial fat oxidation and respiratory chain-related enzyme activities increased; however, cold tolerance did not change, and cardiolipin content and composition also did not change noticeably.[1]

This shows that different indicators can move in different directions. Improved exercise performance cannot be directly written as all energy stress being resolved; unchanged lipid amount in the membrane also cannot be used to deny functional changes.

If the amount does not change, why can function still change?

It is like a machine whose number of parts has not changed, but the parts fit together better, so operation may also improve. The authors propose that SS-31 may help fat oxidation-related enzymes and the respiratory chain work together more stably.

This is a mechanistic explanation consistent with the results and still needs further experimental testing. Responses in patient-derived cells may be affected by genotype, which also suggests that different defects cannot be treated as all the same.

What inspiration does this offer for follow-up experiments?

When designing mitochondrial research, membrane lipid composition, enzyme activity, energy supply, and overall function can be measured separately. If only one indicator is measured, it is hard to know at which level the improvement occurs.

This article provides preclinical evidence related to TFP deficiency. It does not prove that SS-31 can correct genetic defects, nor can it be generalized from this into a universal treatment for all fatigue or mitochondrial diseases.

Can results from fibroblasts represent all organs?

These cells are easy to culture and test, but tissues such as muscle, heart, and nerve have different tasks. Using patient-derived cells makes the model closer to a specific genetic background, but it does not mean it includes responses from all organs.

When reproducing experiments, cell source, genotype, and culture conditions should be recorded. If cells from different sources respond differently, this may be information worth continuing to study and should not simply be averaged into one universal effect.

Remember this point when reading this article

Unchanged cardiolipin content and improved function can both be true at the same time; endurance and cold tolerance also need to be looked at separately.

Related research materials

View materials and analytical information

The SS-31 catalog materials can be used to check the form of mitochondrial-related research materials and analytical documents. Patient cell experiments are still not the same as verification of efficacy in humans.

Mitochondrial function research

SS-31 · Elamipretide

View material form and analytical information, and confirm specifications according to the project.

View product information →

Original materials

Material date: 2026-01-01 · Genetically defective mice + patient-derived cells

Elamipretide Improves Mitochondrial Function in Mitochondrial Trifunctional Protein-Deficient Mice and Human Fibroblasts. ↗

Paper DOI: 10.1002/jimd.70132 ↗

Un peptide qui se lie à la cardiolipine sur la membrane mitochondriale doit-il nécessairement augmenter la quantité de cardiolipine pour agir ?
