D and L describe amino-acid stereochemistry: a difference in three-dimensional arrangement. Left and right hands are a helpful analogy. They have the same fingers, but are not interchangeable in a fitted glove. For peptide ordering, state the arrangement of each relevant residue rather than relying on a short product name.<\/p>\n

An all-D peptide keeps the sequence order<\/h2>\n

As an invented three-residue example, start with L-Ala\u2013L-Phe\u2013L-Lys, written from N to C. The corresponding all-D sequence is D-Ala\u2013D-Phe\u2013D-Lys. The letters stay in the same order, while the specified residue configurations change. This is an ordering illustration, not a tested functional peptide.<\/p>\n

Retro-inverso also reverses the order<\/h2>\n

For that same example, a retro-inverso design is D-Lys\u2013D-Phe\u2013D-Ala, again written from N to C. Do not confuse it with the all-D sequence above. Specify terminal chemistry too. Neither the shorthand nor the hand analogy establishes equivalent binding, stability or biological activity.<\/p>\n

Make the supplier\u2019s notation explicit<\/h2>\n

Bachem\u2019s FAQ provides examples with D-residues explicitly marked in three-letter notation. Your supplier may also use a one-letter convention, but confirm it in writing. Mark each D-residue by position, especially in a mixed D\/L chain. Glycine is achiral and does not have a D\/L pair in this sense.<\/p>\n

Check what the analytical package can distinguish<\/h2>\n

D and L forms have the same molecular mass. A mass result alone therefore cannot prove that the intended stereochemistry was supplied. Ask what records or suitable analytical evidence support the specified configuration. Send a numbered sequence and a structural reference with the enquiry; feasibility and document availability need confirmation.<\/p>\n
