\u201cAdd biotin\u201d or \u201cadd a fluorescent label\u201d sounds like a complete request. It leaves important choices open: which label, where it attaches and whether a spacer sits between the label and peptide. A small drawing can prevent a long round of clarification.<\/p>\n

The same label can sit in different places<\/h2>\n

JPT\u2019s modification guidance describes terminal and side-chain labeling options. For a teaching example, imagine a peptide with one lysine that can be used as an attachment site. N-terminal labeling and labeling that lysine side chain describe two different designs. Whether either design is feasible or suitable requires project review.<\/p>\n

What is a linker?<\/h2>\n

Think of a linker as a short connector between the peptide and tag. Write its chemical identity rather than just requesting a \u201clong linker.\u201d If the published material uses a particular connector, include that description or structure. Do not assume that adding distance will preserve binding or improve a signal.<\/p>\n

Count what is included in the final molecule<\/h2>\n

A modified peptide includes the chain, label, linker if present, and the specified terminal chemistry. Use the molecular weight of that final structure for mass-to-moles calculations. The exact mass change depends on the attachment chemistry; adding the molecular weights of free starting reagents can give the wrong answer.<\/p>\n

Send a design that can be checked<\/h2>\n

Include the full sequence, exact label, attachment residue or terminus, linker identity and remaining terminal groups. For fluorescent designs, identify the dye and the instrument or assay requirement. Ask how identity, purity and labeling position will be checked, and list any functional testing you need separately.<\/p>\n
