A peptide library averages 91% purity. What is the weakest sequence?

An average can hide the one sample that causes trouble. When ordering a peptide library, a mean purity value is useful background, but it cannot replace the result for each sequence.

Five numbers show the problem

Consider a hypothetical library with individual purities of 99%, 99%, 99%, 99% and 59%. Their simple arithmetic mean is 91%: (99 + 99 + 99 + 99 + 59) ÷ 5. Yet one peptide is only 59% pure. This is a teaching example, not a supplier’s batch data. It also is not a measurement of a pooled mixture.

A per-peptide requirement answers a different question

“Average purity at least 90%” and “every peptide at least 90%” are different specifications. JPT’s peptide-quality guidance explicitly discusses why an average can conceal low-purity individual peptides. A requirement for every sequence should say what happens when one fails, rather than leaving that decision until delivery.

Match every result to an identifier

A useful library report links each sequence to its own identifier, analytical result and delivered amount. Check that the identifiers on the plate or tubes match the document. Missing or failed sequences should be visible. An attractive summary chart is less useful if you cannot find the result for the peptide used in a particular experiment.

Agree on exceptions before synthesis

Ask whether difficult sequences will be reworked, reported as exceptions, omitted or supplied under an agreed lower specification. Changes to sequences should require explicit agreement. Also distinguish individual samples from pooled delivery: a pool creates additional questions about mixing amounts and traceability. The right specification is the one that supports your assay, not simply the largest percentage in the quotation.

Next step with NHD

The related product cards below identify catalogue peptide families. They do not confirm availability of biotinylated, fluorescent, isotope-labelled or library versions. For a modified material, specify the exact design in a custom inquiry.

Start with the Peptide library ordering guide. For a quotation, send NHD your sequence or product name, quantity and required documents. Confirm the exact form, feasibility, available testing and delivery terms before ordering.
