Peptide salts: a theoretical counterion ratio is not a batch measurement

By NHD Technical TeamPublished
Editorial comic illustration: Peptide Salt Ratios. Conceptual cover, not experimental imagery.

A peptide may be supplied as a salt with an associated counterion. A proposed ratio can be useful for planning, but the calculated ratio does not replace an actual compositional result.

What a ratio tells you

A stated molecular model describes what is being assumed. An analytical result describes what was measured under a method. Depending on the sample, water, excess acid and other components can affect the observed composition. A catalogue name alone does not quantify them.

A labeled teaching example

Take a hypothetical peptide with a neutral mass of 1,000 g/mol and exactly one acetate counterion represented as 59 g/mol. Ignoring other components, the peptide fraction in that model is 1,000 ÷ 1,059, or about 94.4%. This intentionally simplified calculation is not a specification or a prediction for any listed product.

Why the real powder may differ

Different charge states, counterion content and moisture can change the mass balance. Also, a salt-form name and a residual-TFA measurement answer different questions. A material described as an acetate salt may still need a separate residual-TFA result when the project requires it.

Ask for the form and the measurement

State the desired salt form and any specific counterion limits separately. Request the measurement method and reporting units where available. Avoid estimating a batch’s content from sequence alone when the experiment depends on a known quantity.
