Cagrilintide and Semaglutide: reading the REDEFINE 1 combination result

REDEFINE 1 tested cagrilintide and semaglutide together, each ingredient separately, and placebo. Comparing these four groups explains the result more clearly than adding numbers from unrelated trials. This guide shows which preparation, participants and analysis produced the finding.

What is the combination trying to do?

Cagrilintide is an amylin analogue, while semaglutide acts through the GLP-1 receptor. These are different signaling pathways relevant to appetite and metabolism. A combination study asks whether using the two together produces a different result from either alone. That question is more useful than assuming that putting two promising molecules together must double their effect.

Who took part?

The 2025 REDEFINE 1 trial included 3,417 adults with overweight or obesity without diabetes. It lasted 68 weeks and included combination, semaglutide-alone, cagrilintide-alone and placebo groups. All groups also received lifestyle intervention. The original paper is the source for the comparison.

Use numbers from the same analysis

Under the treatment-policy analysis, estimated mean body-weight changes were −20.4% for the combination, −14.9% for semaglutide, −11.5% for cagrilintide and −3.0% for placebo. This analysis addresses the assigned-treatment strategy, including what happens when treatment is stopped. These four values can be compared because they use the same framework. A headline using an adherence-assuming estimate answers a different statistical question.

Combination results are not simple addition

The two single-component reductions should not be added to predict the combination result. Biological responses can overlap, and the same endpoint has practical limits. The trial directly measured the combined intervention instead. It also recorded gastrointestinal adverse events. A complete comparison considers how much weight changed and how well the intervention was tolerated.

Do not mix REDEFINE 1 and REDEFINE 2

REDEFINE 1 studied people without diabetes. REDEFINE 2 addressed people with type 2 diabetes. A different starting population can produce a different average result. Our REDEFINE 2 explanation covers that separate trial. Neither study is a shortcut to predicting a particular person’s response.

Find the related NHD catalogue records

See the Cagrilintide page and the Cagrilintide–Semaglutide research-blend page. The catalogue blend is not established as equivalent to the clinical trial formulation. A formulation-research enquiry should state the required components, ratio and analytical testing, rather than relying on the combination name alone.

Keep the combination and its ingredients separate

Check What it tells you
Which intervention? CagriSema is a clinical combination studied with a defined formulation and protocol.
Which catalog record? Cagrilintide alone and a cagrilintide–semaglutide blend have separate identities and specifications.
Which conclusion? A combination result cannot establish the same result for either ingredient alone or for a different blend.

For laboratory procurement, review the related NHD catalog material and confirm the selected variant, amount basis and available records. Send a research enquiry with those requirements; the cited experiment is not a test of an NHD lot.
