A change in color, fluorescence or light output has several possible causes. Check cell number and assay interference before assigning the change to the pathway you want to study.

A readout is a marker, not the cell count itself
Common assays use metabolic activity or enzyme reactions associated with living cells to produce a signal. Cell number, each cell’s condition and effects on the reagent reaction can all alter that signal. Calling a decrease pathway inhibition skips an important check.
Separate at least three questions
Did the treatment change the target response? Were cells damaged or reduced in number? Did the material affect detection directly? More than one can occur. Cross-checking methods based on different principles is more informative than continually adding replicate wells to the same assay.

What a cell-free control can reveal
Adding the test material to the same medium and detection reagents without cells can help identify color, fluorescence or chemical interference from the material itself. It cannot answer every intracellular question, but it can prevent obvious reagent interference from being mistaken for a biological effect.
A calculation that is easy to misread
Hypothetical example: total signal falls by 40% while the number of living cells also falls by 40%. In the simplest linear case, average signal per living cell may not have fallen. This calculation is a reminder to obtain cell-number information, not a recommendation to divide every readout by live-cell number without validation.
Match KPV and LL-37 readouts to the research question
For inflammation-related responses, microbes or cell behavior, state the main endpoint and how cell condition will be checked. Describing your purpose and detection platform when enquiring about material helps identify identity, purity and interference checks to discuss. A catalog name does not rule out assay interference.
